Bromelain is a protease enzyme obtained from pineapple stems that consists of a mixture of various thiol endopeptidases and other components such as phosphatase, glucosidase, peroxidase, cellulase, echarase and several protease inhibitors. It works by selectively inhibiting the biosynthesis of pro-inflammatory prostaglandins, and also has analgesic, anticancer and proapoptotic properties. Bromelain has potential therapeutic efficacy in the treatment of conditions such as angina, bronchitis, sinusitis, surgical trauma, and osteoarthritis. It is considered a safe food supplement.
Chymotrypsin (EC 3.4.21.1) is a digestive enzyme that promotes proteolysis, that is, the breakdown of proteins and polypeptides. It is a serine protease synthesized in the pancreas and is an important component of pancreatic juice. Like most proteolytic enzymes, chymotrypsin is activated from its inactive zymogen precursor, chymotrypsinogen, in the presence of trypsin. Chymotrypsin is the most abundant pancreatic protease, which accounts for up to 10-20% of the total proteins synthesized by the exocrine pancreas. Chymotrypsin contains both a catalytic triad and an oxyanion hole, and the tertiary structure of chymotrypsin is similar to that of trypsin.

How Wobenzym N works
Chymotrypsin is synthesized by pancreatic acinar cells as an inactive precursor, chymotrypsinogen, which is secreted into the duodenum and activated by trypsin-induced cleavage. It also causes its own activation by cleaving essential amino acid residues in the oxyanion cleft to produce α-chymotrypsin, which is a more stable form than π-chymotrypsin. Residues His-57, Asp-102, and Ser-195 form the catalytic triad, while residues 189-195, 214-220, and 225-228 form the primary substrate binding pocket, called the S1 binding pocket. Residue 189 in the polar serine residue at the bottom of the S1 binding pocket. Chymotrypsin prefers aromatic residues such as phenylalanine, tyrosine, and tryptophan, but can hydrolyze other bonds in peptides more slowly.
Pancrelipase is used to compensate for pancreatic enzyme deficiency. As mentioned above, pancrelipase is a mixture of lipase, protease and amylase that can break down fats, proteins and starches in the small intestine. When applied topically, papain elicits an allergen-like inflammatory response by recruiting neutrophils, mast cells, and CD3+ cells and inducing a TH2-dependent antibody response.In vitropapain treatment disrupted the tight junctions of primary human keratinocytes that maintain epithelial barrier integrity. These tight junction proteins include zonula occludens-1, claudin-4, and occludin. Papain is believed to induce allergic reactions via TLR4 activation, leading to an increase in neutrophils, CD3+ cells, mast cells, and CCL8-positive cells.
